Route 376: Route is roughly 6 miles, Google Maps says the most direct route between Beckton Station and Newham Town Hall (shared by the 101/474) is only 1.3 miles.

Route 80 - runs in a horseshoe shape between Belmont Prisons & Hackbridge despite there being a much direct route between the two places. Google Maps quotes 4 miles for the quickest route whilst the 80 is 9 miles long.
Route 157 - takes a very round the houses route between Morden & Crystal Palace. Google Maps quotes 7.3 miles for the quickest route whilst the 157 is 12 miles long.
Route 412 - direct route between Purley & West Croydon via Brighton Road yet the 412 runs right out to Selsdon & Sanderstead before heading inwards towards Purley. Google Maps quotes 3.4 miles for the quickest route whilst the 412 is 7 miles long.

163 from Wimbledon to Morden, horseshoe route via Raynes Park and Lower Morden... compared to the more direct 93 via South Wimbledon
381 and C10 is rather circuitous between Surrey Quays and Jamaica Road (via Rotherhite) compared to the more direct 47 and 188
As already mentioned by member 'Someone' I think the most circuitous one I know of in south London is the G1. After five minutes within the St Georges Hospital complex alone a driver would often return to Garrate Lane forgetting which direction they are supposed to go.
When I drove it briefly under Limebourne I was too late in realising I went the wrong way when I turned right into Garrate Lane when I should have turned left towards Battersea. I managed to quickly turn it around instead of doing so at Tooting Broadway
Also, if a passenger gets on the G1 from Clapham Junction asking if it goes to St Leonard's Church I used to tell them,
"yes! but the 319 gets there in half the time"
or
"yes, but this one goes round the houses in a big way"
I will never forget when one rather jolly cockney passenger gave it a go before eventually coming up to me and said, "F__k me you weren't kidding when it said it went round the houses, I've flipping grown a beard since Clapham Junction"

The 171 is a great example. From Brockley to Elephant & Castle/Waterloo the 172 serves a much quicker route.

Route 199 is a bit circuitous as the route and the 47 run from the same place in which they run together towards Lewisham. The 199 then separates from the 47 via Greenwich, then rejoins the 47 at Deptford, then separates from the 47 again via Pepys Estate, then rejoins with the 47 at Surrey Quays Station, then separates from the route via Surrey Quays Road before meeting the 47 again at Canada Water.
Route D3 is circuitous around the Crossharbour/Island Gardens area in which it separates from the 135 at Crossharbour and runs via Marsh Wall, Isle of Dogs and Island Gardens Station, then runs via Mudchute Station along with the 135, then turns into Spindrift Avenue before meeting the 135, D7 and N550 at Manchester Road.
Routes 472 and N1 are circuitous around the Crossway area in which they do a full loop around there before terminating at Titmuss Avenue and Thamesmead Town Centre.

Surely the 386 is in the running? It seems to take the most circuitous route it can between any two points - you can easily walk from Blackheath Village to the Standard quicker than doing it on a 386, and it's no more direct from there to Woolwich. Similarly the R3 gives you a nice tour of the environs of Orpington and Petts Wood, but get on it wanting the Princess Royal Hospital in a hurry and you're in for a shock.

The 152 and 200 parallel each other from Mitcham through Colliers Wood and Raynes Park, the 200 much more circuitous in comparison (via Phipps Bridge, Haydens Road and Wimbledon Hill).

I very nearly posted the 386, haven't forgotten the time I made the mistake of taking it between Greenwich and Woolwich

381 and C10 is rather circuitous between Surrey Quays and Jamaica Road (via Rotherhite) compared to the more direct 47 and 188 I'd add that Route 381 also takes a zig-zag route from the Old Kent Road to Surrey Quays via The Blue, also the same for the P12 between Ilderton Road/South Bermondsey Station and Surrey Quays via the Blue and Jamaica Road.
