One of the more interesting examples is the 229 and 269.
The RM operated (previously RT) 229 ran from Farnborough to Woolwich but was cut back to Bexleyheath Garage with a new OPO route 269 between Sidcup and Woolwich in 1977. Subsequent changes over the years mean the routes have swapped ends with the 269 now running between Bromley and Bexleyheath and the 229 between Sidcup and Thamesmead.

Route 21 cut back from Foots Cray to Lewisham. Replaced with new 321 route. - P3 renumbered 343 and extended to terminate at City Hall - Route 205 extended to Bow Church(?) when Stagecoach took over. During the Olympics in 2012, routes 129, 177, 180, 188, 199, 286, 386 and N1 (heading towards Deptford or East Greenwich) were re-routed via Norman Road due to the one-lane closure of Greenwich High Road. I remember that. There was complete chaos for a good week from when it started.

Route 165 Withdrawn from Havering Park and Romford and replace by 365 (1990 or 1991). Mard**e Estate section replaced by 365 (1988).
Route 252 Withdrawn from Gidea Park Station and Hornchurch also School journeys between Collier Row and Chase Cross replaced by 651.
Route 248 Withdrawn from Romford Market and Romford St Edwards School and replaced by 686.
Route 174 Withdrawn from Noak Hill and Harold Hill re-routed to Dagenam Park Square replace withdrawn route 374.

133 - cut back from Tooting Broadway to Streatham, St. Leonard's Church (2003) - this was to make way for the newly created 333. Then in 2007 the N133 was also cut back from Tooting Broadway to St Leonard's and rerouted to Mitcham. The 333 was also rerouted between Brixton and Kennington to go via Stockwell Road and Clapham Road rather than Brixton Road.

- 131 cut from West Molesey to Kingston, replaced by the 411 - 10 cut from Archway to King's Cross, replaced by the 390 - 276 diverted away from Carpenter's Road due to the Olympics - 276 withdrawn between East Beckton & Newham Hospital - 44 diverted from Vauxhall to Victoria (?) - 142 diverted to Brent Cross instead of Kilburn Park (replaced by 32?) - 14A (early 1990s) diverted from Warren Street to Trafalgar Square and renumbered 91 - 74 curtailed to Putney Garage from Roehampton, replaced by 430 The 131 used to run beyond Molesey to Walton (Manor Rd). The Kingston - Wimbledon section was a trolleybus replacement 604? Therefore the current 131 literally only does few metres in Kingston of its former route (and that is debateable as road layout has changed) but its safe to say its over 99% rerouted (and I assume this thread excludes route numbers reused in another part of London)

50 - northbound terminus used to be Stockwell, but via Brixton during the 80s
70 - completely different route, what is basically now the 381
95 - completely different route, what is basically now the 333
181 - completely different route, what is basically now the 333 (streatham to tooting Broadway) and (155 tooting Broadway to Stockwell). Not sure where it went beyond stockwell

Route 253 cut back from Aldgate to Hackney. New Route 254 created to partially replace section lost.

The P13 has also had numerous re-routings in the East Dulwich area, diverted away from Grove Hill Road to serve Pytchley Road and was also re-routed away from Lordship Lane between Whateley Road and Melford Road to serve Underhill Road.
Route 78. Extended to Forest Hill in part to replace capacity between FH and Dulwich library from Route 12, then curtailed to Peckham Rye and then extended to Nunhead.
185 was curtailed from Greenwich to Lewisham in the 80s.
Route P4 was re-routed from Lordship Lane and Court Lane to serve Dulwich Common and College Road in the early 90s.
Route P12, extended north from Peckham Bus Station to Surrey Quays using the former P13 route and south to Brockley Rise via Brenchley Gardens.
Route 63, cut between Honor Oak and Crystal Palace, replaced by Route 363.
Route 37, extended from Putney Station to Putney Heath.
