Here's a couple I've conjured in the past couple days:
Route 370 is the only route to serve the same train station three times: Upminster Station, Upminster Station Forecourt and Upminster Station/ St Lawrence Road.
There are now four routes running in different directions between Canada Water and Waterloo: the 1, 188, 381 and C10. I thought someone already mentioned this fact but didn't come up in a search on this thread.

The multiple routes between two points thing was covered a while ago - I think someone came up with an example of 5 or 6 different routes between two points, but can't remember the details now.

That is probably the most, certainly can't think of more. I can think of three near me that have almost completely different routings for their entire journeys - 249, 322 & 417 between Clapham Common & Crystal Palace largely stay away from each other between the two places except for the 417 sharing three stops westbound with the 249 (2 eastbound with the 249) on Crown Lane and three stops in Palace, the 322 only shares four stops in Palace with the 417 and just one stop at Clapham Common with the 417 & the 249 only shares three stops with the 322 at Palace and just the first stop at Clapham Common for both the 249 & 322.

My favourite route to get to Lewisham from Woolwich (before they changed the dxmn speed limit) was the 122.

If multiple routes following the same route are allowed, then Kingston to Surbiton is a very interesting case.
Just including TfL bus routes, there are 4 different routes: 71/281/K2/K3, 465, K1 and K4. However, if non-TfL routes can be included, then there are actually 6 different routings, as the 515 uses Kingston Hall Road and High Street instead of Brook Street, and the Kingston University buses use Wheatfield Way instead of Eden Street.

Brixton to Elephant (35, 133, 196, 333 and P5) That's probably the only other example, although that depends on whether duplicates are allowed, since the 35/45 and 133/415 have the same routings. 4 buses Uxbridge - West Drayton - all with (in some cases slightly) different routings: 222/U1/U3/U5

Elephant to Peckham is another one and this one. They come in 2's 136 and 343 on one part 12 and 171 on another and the 63 and 363. I think I mentioned this a while back.

Just including TfL bus routes, there are 4 different routes: 71/281/K2/K3, 465, K1 and K4. However, if non-TfL routes can be included, then there are actually 6 different routings, as the 515 uses Kingston Hall Road and High Street instead of Brook Street, and the Kingston University buses use Wheatfield Way instead of Eden Street. That's jogged my memory. Some years ago I worked out there were 8 different routeings between Kingston and Esher. This involved a number of Surrey routes that no longer exist, including school routes. Haven't kept up with the various changes round there but I doubt it's anywhere near 8 routeings now.
