Mine were the 258 from Harrow town centre to Watford High Street (1 hour). Big mistake not taking the train to Watford.
Also the 95 from Greenford Broadway to White City was around 1 hour.
Both were a few weeks before Xmas and involved heavy traffic
Although not the longest, one time the 140 from Hayes and Harlington to Barnhill High School was very slow and the bus hardly moved soon after Uxbridge Road

2 hours 20 min on the 25 eastbound, from Oxford Circus to ilford. This was the last week before it was hacked back to Holborn Circus
Think have done a bunch of routes end to end between 90 - 120 min (x26, 123, 266 were all 90 min or so). Also did 2 hours on a 142 from Brent Cross to Watford when it was thrown into disarray due to 1 temp traffic light

First X53 when it ran from Weymouth to Exeter, a three-hour journey which increased to 3 hours 15 minutes when it was rerouted via Axminster. Did that a few times.
Stagecoach still has some long runs out of Exeter - the 5B to Barnstaple and the 6 to Bude both take more than 2 hours. Similarly Go Cornwall Bus 11 Plymouth-Padstow and 12 Plymouth-Bude.

I have had a few bad journeys on the 5 of about 90 mins.
I really old example was back in 1995 (yes 1995) I was at College at Edgware Road and thought it would be a good idea to catch a 15 back to east London during the evening peak! was over 2 hours! - Never did this again!

Done the whole X26 once which took over 2 hours. Also did 18 from Euston to Sudbury which took almost 2 hours. A couple of non-TfL ones I did was the 724 between Hertford and Watford & 300/301? (forgot which one I took) between Stevenage and Hemel Hempstead which both took over 2 hours

Was driving non stop, then reached driving hour limit of 5 hours 30 mins, sat on the bus for 30 mins, continued to depot then eventually took break.

Outside of London, the 702 probably was the longest and it was coming from Reading to Victoria due to heavy traffic at Legoland - it was timetabled at 2h, 31 minutes but we were sitting at Legoland in traffic for a good while so probably closer to 3 minutes. Other notable mentions was the 102 which took 2h 21 minutes from Dover to Rye and that's with good conditions.

More than an hour on a 232 Streetlite from Turnpike Lane to Neasden in April... had to go to court at Willesden, only to find it adjourned and had to return in August had a long wait at Henley's Corner to "Regulate the Service", only to find the place closed due to COVID-19. Back in the day when First ran the 25, I took a ride on then new VN36139 to see what a Voith B9TL sounded like from Bank to Ilford. Slow driver and heavy traffic spoiled that for me Apart from doing the entire 243, 258, 370 routes, it's more short hops these days Have spent over an hour waiting for a 65 in Richmond when Armchair ran the route with Atlanteans (though this was back in 1995)

Despite all the routes I use such as the 25 and 205, the route that holds my record is the 368 spent over two hours on a bus once when someone jumped off a bridge on the A13 and sent all of East London into total gridlock, the whole A13 was shut from Dagenham Asda to the Beckton roundabout and the A406 was a standstill up to North London.

Once spent about 2 and half hours getting from Kingston to Raynes Park on a 131. This was just after 7pm on a Friday night in the rain. Used to be fairly regular on the 131 but I’d never experienced a trip like that even with the traditional notoriously traffic heavy Fountain Roundabout approach.

I think I’d say Peterborough to Lewisham on Go-Ahead’s RM 9 back in the 90s. I don’t know exactly how long it was but it says much for the Routemaster that I slept like a baby for virtually the whole journey 🤣

For me, it was an X26 journey from Heathrow to West Croydon taking two hours.
Other than that, I was once stuck on a 162 for an hour and 10 minutes between Eltham and Bromley a couple of years ago due to some roadworks near Chislehurst War Memorial. It came as no surprise that the bus was terminated at Bromley South as a result.

Many occasions I have spent 90+ min on a 607, Can remember 1 journey taking well over 2 hours once, roadworks in Acton followed by taking over an hour to get through Southall lead to the TNL getting a good thrashing between Trinity Road and Uxbridge. Spent just shy of 3 hours on an X26 once! That was pure torture! Don't think my bladder will ever forgive me for that! Just a bad combination of roadworks and traffic. Not being so clued up on bus routes in South/SW London, I had to stick with it. The driver giving the poor Citaro the hiding of it's life on the final run into Croydon kinda made up for it! Caught L1/DMP42401 on the U3 once, took an hour to travel the 3 stops from Uxbridge Station to Vine Street. Can remember seeing a mates GF waiting for her 331/U9 at the Uxbridge High Street stop, for about 20min we were just looking and laughing at each other shrugging our shoulders, before she called me up and was like 'This is embarrassing' 90 min after boarding, I finally got off at Appletree Ave. I should of bailed out and walked, but it was winter and cold... One from last year, caught the Greenline 703, won't do that again! Having posted about it in the Reading Buses thread, over 2 hours to get to Bracknell and 2.5 hours to get back, totally unacceptable! bus was just crawling along for no reason. My GF still gets wound up now when I mention that day! Not a bus journey but as a young kid, when we used to visit our Grandparents in Southport, we'd get the National Express from Victoria. 5.5 hours felt like torture! Can remember one time, it felt like we was on the go for ages, when I piped up to my dad 'Dad, I can see the sea' He looked at me and laughed, we hadn't even reached Golders Green..... After that we started taking the West Coast Mainline!

X26 seems to be a common one, also fell victim to its immense journey time.
I also fell victim to traffic on Knightsbridge, on a packed 14, with people filling up along the staircase, myself included, practically inescapable. It took about an hour to get from Knightsbridge, Harrods to Trocadero, Haymarket. I felt incredibly bored and was on the verge of falling asleep.
Over a decade ago, back when Metroline operated the 24 (originally), I was with my parents, trying to get to Victoria, but due to the journey being so long, and it being late at night, my parents fell asleep, and we just went to the end, to Pimlico. I woke them up at the end of the journey, and after getting off the bus, we discovered that people were throwing things off buildings, but not at us directly, thankfully. We eventually got back to Victoria and then took a train home.

Like others here have said the longest singular I have been in London end to end is the X26 where I actually have done it from Croydon to Heathrow and vice versa from/catching flights not just for bus enthusiast reasons. Due to it's directness. There has been times where being on the route where traffic jams have happened at Worcester Park, New Malden and Kingston when travelling on this route so I know that has pushed my time being on it. Also have had a situation being on the route when the Carshalton Fireworks happened which was a unique experience, even got curtailed at Wallington Green that time from Heathrow, must of been on the bus for more then 3 hours.
I've done various trips just being on just a bus when younger for enthusiast reasons one I can think off the top of my head is from Highbury and Islington 263 to Barnet, 307 to Ponders End, 349 to Edmonton Green, 149 to Liverpool Street, 133 to Elephant and Castle, 468 to Croydon along them lines. So that's probably the longest I've been on just a bus in one chain but then I would of had to get off so not sure if that would count.
Longest singular bus route I've been on is probably the X62 from Hull to Leeds which even though was a bus with DD Buses, ran by Stagecoach East Midlands actually sold Megabus tickets which were cheaper then the bus ones and also had Megabus drivers and a Coach like timetable. This route even goes on the M62 which is a very unique experience to be on. If not that then X47 to York from Hull which is a longer version of the X46 covering the same route going through a different route through more villages and back streets. X62 is like 2 hours and a half, X46/47 is like 2 hours. Also way less traffic on these routes compared to London especially X62.
