If London was deregulated I'd be looking for a busy corridor that and make routes that are not too long thus manageable within drivers hours. As I know south London best, I'll go for these, stopping at all stops.
Lewisham - New Cross - Old Kent Road - Elephant then alternating to Waterloo - Holborn - Oxford Circus or London Bridge - Bank - Cannon Street - Holborn. As an amendment on Sat/Suns and after 20.00 weekdays route 53B version will terminate at London Bridge as it deregulated and I am only interested in profit. During the night buses after Elephant operate as route 12 after Elephant to Oxford Circus. Numbered 53A, 53B and 53N
Putney - Wandsworth - Clapham Junction - Vauxhall - Victoria - Westminster - Waterloo - London Bridge numbered 77A
I'd expect lots of competition!

Two more I can think off are:
Pollards Hill via 463 to Eastfields, up Locks Lane to Figgs Marsh, via 270 to Garrate Lane/Burntwood Lane, full length of Burntwood Lane then via 319 to Clapham Junction.
New direct link Pollards Hill to Tooting, (which I believe was meant to have been done with the 352 back in the 90s) and re-established link Mitcham to Clapham Junction, lost when the 219 was shortened/diverted from Mitcham to St George's Hospital.
Other route would be Canada Water or Surrey Quay to Piccadilly Circus via 47 to Monument and 15 to Piccadilly, giving London Bridge a direct bus link with the west end.
Was actually shocked to realise there's no such direct bus link, despite the Jubilee Line alternative.

Do people think its fairly likely that the old established coaching routes and roman roads would be able to sustain a commercial service? Edgware Road, Harrow Road, Uxbridge Road, etc etc.

The idea is for Longer distance ridership ie commuters and people from further afield needing to get to St Thomas's plenty of other alternatives in Streatham/Brixton Very much the same logic as the X68 skipping Elephant and Camberwell In addition to that, it should skip West Norwood too in the mornings. Usually the buses are jam packed by the time they reach the 'Norwood Crown Point' bus stop, let alone West Norwood!

I have to say that I rather like that route. I'd even be tempted to run it down Pole Hill Rd and then straight into Uxbridge. It would be long but the links are potentially very useful indeed. I'd even be tempted to run it 24 hours given the very high frequency of services to Shepherds Bush for connections. Would you withdraw the U7 service or keep it? Would the 195 be affected given there is overlap through Hayes and Southall?

I would keep the U7 for local links to Hillingdon Hospital and plus the U7 and 95 would have different routings to Uxbridge but I would withdraw the 195 to stop the overlap with the 95 and extend the H32 to Brentford via the withdrawn section of 195 between Southall and Brentford to provide a extra capacity and to retain local links to Ealing Hospital.

You could, instead of Uxbridge Road direct, take it down Lees Road, then follow the U4/U7 to Uxbridge, that way you retain the vast majority of links to Hillingdon Hospital and allow the U7 to be withdrawn. Its a very long route though either way, meandering either side of the Uxbridge Road. The 207 ran mostly split into two sections for many years, and that takes a more direct route. I've had to catch the U7 before a few times, its not a great bus to wait for at Uxbridge, was often longer than a half hour wait aswell so a high frequency replacement would presumably be welcome
