I KNOW it's not bus related but the ultimate 'opposite direction' must be Exeter to London? Waterloo and Paddington trains leave in opposite directions I thought Edinburgh to London might be the ultimate. West coast and east coast trains leave Edinburgh in opposite directions towards London

340 and H18 at Harrow Weald towards Harrow 340 and H10 at Wealdstone towards Harrow
Not the same terminus but at Preston Road, 223 and 79/204 head to Wembley Central in opposite directions

Virgin west coast has a hourly London to Scotland service via Birmingham that alternates between Glasgow and Edinburgh

Virgin west coast has a hourly London to Scotland service via Birmingham that alternates between Glasgow and Edinburgh Indeed, a journey planner enquiry "London to Edinburgh" (or v.v.) will show both routes, and people quite often select the London Euston-Edinburgh one if the advance fares are cheaper, or they want to travel up one way and back the other to see different scenery.

This is different to my OP but I wonder if the following situation is unusual. Others will tell us I'm sure. Two routes pass in opposite directions going to DIFFERENT termini, but then the same two VEHICLES pass in opposite directions again further along their route. 499 and 256 pass in Hilldene Avenue in opposite directions , 499 towards Romford, 256 towards Noak Hill, then five minutes later the same vehicles pass in opposite directions in Noak Hill Road.

104 & 238 to Stratford and 5 & 300 to Canning Town pass each other in opposite directions in East Ham 8 & 205 to Bow Church both pass each other in Liverpool Street To add to that, even though they do not terminate at the same place, 252 and 247/294 pass each other in Collier Row going in opposite directions towards Romford

Here are some examples I can think of although I don’t know if they can meet twice due to timetabling. 182/186/258 with H9/10 at Wealdstone High Street and again at Station Road, Harrow Town Centre 182/258 with H18/19 at Harrow Weald and again at Station Road, Harrow Town Centre 224 and 487 at Acton Lane between Central Middlesex Hospital and Harlesden station and again very briefly in Alperton between station/garage and Sainsbury’s. This ones a bit of a cheat because of the 224 double run in Alperton. 142 and 324 in Stanmore between station and broadway and at Brent Cross 114 and H9/10 could possibly meet twice at several points along their routes: Between Kenton library and Harrow High school (although H9/10 has a couple of deviations in this section) Along College road east of Harrow bus station On Bessborough Road between College Road and Lascelles Ave Between Northolt Road/Shaftesbury Ave in South Harrow and Eastcote Lane/Kings Road

Rye Lane, Peckham - the 343 and P13 go in opposite directions towards New Cross Gate.
As for trains there are some stops in London where false destinations are not used on services departing in opposite directions towards London - Hounslow and Strawberry Hill towards London Waterloo. Ramsgate is another example, trains towards St Pancras depart in opposite directions.
